AB OBJECTIVE: We have shown that MT-4 cells survived the infection with
vif-, vpr-, or vpu-mutant HIV-1, but not with wild-type or nef-mutant
HIV-1. However, increasing persistent infection in the cells were
observed by serial passage of wild-type HIV-1, but not by serial passage
of nef-mutant HIV-1 even after 50 passages. Here, we focused on the
naturally occurring genome mutations in the serial passages of wild-type
HIV-1. METHODS: Transfection products of infectious molecular clone
pNL432 and pNL-Xh (inserting a frame-shift mutation of 4 bases at the
Xho I site in pNL432) were used as wild-type and nef-mutant HIV-1,
respectively. Serial passages of these viruses were repeated up to 50
times and were designated as wt-1 to wt-50 and nef(-)-1 to nef(-)-50,
respectively. The PCR products in infected cells were cloned, then
sequenced. RESULTS: All DNA clones derived from PCR products of the
cells persistently infected with wt-20 and wt-50 contained termination
codon in vpr. Substitution in vif gene was also observed in some DNA
clones. Especially, a large deletion within vif to vpr genes was
identified in some DNA clones. These structures were also identified in
the M10 cells acutely infected with serial passages of wild-type, but
not nef-mutant HIV-1. DISCUSSION AND CONCLUSIONS: The vif and vpr gene
mutations essential for persistent infection were progressively induced
by serial passage of wild-type HIV-1. The nef gene might be important
for such mutations induced during the serial passage.
